I am involved in the research on horizontal transfer of genetic information via broad-host-range (BHR) plasmids. These mobile genetic elements are important in that they facilitate the inter-species transfer of genes that allow bacteria to live in the presence of environmental stresses such as heavy metals, chlorinated hydrocarbons, and antibiotics.
One part of the research aims at understanding the diversity and evolutionary history of BHR plasmids. This work entails capturing novel BHR plasmids from the environment (industrial, agricultural and hospital sites, waste water treatment facilities), and extracting large amounts of high-quality plasmid DNA. These new plasmids will be sequenced in conjunction with the Joint Genome Institute, and the sequences will be functionally annotated and compared to already known BHR plasmids. More information on the genome sequences of these important mobile elements will not only improve our insight in their diversity and evolutionary history, but it will also allow us to better use their ‘valuable’ traits, such as in bioremediation, and to counter their ‘detrimental’ traits, such as antibiotic resistance.
The second part of the research studies the effects of spatial structure on plasmid transfer and persistence in bacterial populations. Our main question here is: Can plasmids persist in, or even invade, bacterial populations in the absence of selection, and how? I am working with Dr. Haruo Suzuki and Randal Fox on their experiments addressing the effects of spatial structure on plasmid spread on agar plates and in biofilms.
